Introduction: Cutaneous leishmaniasis is a neglected vector-borne parasitic disease prevalent in 92 countries with approximately one million new infections annually. Interactions between vector saliva and the human host alter the response to infection and outcome of disease. Methods: To characterize the human immunological responses developed against saliva of Phlebotomus duboscqi, a Leishmania major (L. major) vector, we repeatedly exposed the arms of 14 healthy U.S volunteers to uninfected P. duboscqi bites. Blood was collected a week after each exposure and used to assess total IgG antibodies against the proteins of P. duboscqi salivary gland homogenate (SGH) and the levels of IFN-gamma and IL-10 from peripheral blood mononuclear cells (PBMCs) stimulated with SGH or recombinant sand fly proteins. We analyzed skin punch biopsies of the human volunteer arms from the insect bite site and control skin site after multiple P. duboscqi exposures (four volunteers) using immunohistochemical staining. Results: A variety of immediate insect bite skin reactions were observed. Late skin reactions to insect bites were characterized by macular hyperpigmentation and/or erythematous papules. Hematoxylin and eosin staining showed moderate mononuclear skin infiltrate with eosinophils in those challenged recently (within 2 months), eosinophils were not seen in biopsies with recall challenge (6 month post bites). An increase in plasma antigen-specific IgG responses to SGH was observed over time. Western Blot results showed strong plasma reactivity to five P. duboscqi salivary proteins. Importantly, volunteers developed a cellular immunity characterized by the secretion of IFN-gamma upon PBMC stimulation with P. duboscqi SGH and recombinant antigens. Discussion: Our results demonstrate that humans mounted a local and systemic immune response against P. duboscqi salivary proteins. Specifically, PduM02/SP15-like and PduM73/adenosine deaminase recombinant salivary proteins triggered a Th1 type immune response that might be considered in future development of a potential Leishmania vaccine.

Mammary neoplasms are common in felines species and represent a significant disease for its unfavorable prognosis. Changes in the blood count and serum biochemical profile of these patients have potential as non-invasive prognostic markers prior to mastectomy, however, they are poorly described in literature. In this study univariate and multivariate analyses were performed using these factors to determine the effect of each parameter on the one-year survival time after the surgical procedure in these animals. The median overall survival (OS) and the disease-free survival (DFS) were 365 and 242 days, respectively. In univariate analysis, values within the reference range of monocyte, platelet and creatinine counts were identified as significant prognostic factors for OS and only creatinine was significant for DFS (P < 0.05). In the multivariate analysis, platelets and mean corpuscular hemoglobin concentration (MCHC) remained independent prognostic factors for OS. The results presented suggest that monocytes, platelets and creatinine may be important non-invasive pre-surgical prognostic markers, and that platelet count and MCHC are independent prognostic markers for feline mammary carcinomas (FMC). The correlation between such alterations is of important relevance for veterinary oncology, and prospective studies are needed to validate their clinical use and that platelet count and MCHC are independent prognostic markers for FMC. The results found in this study can also be studied in human medicine, regarding blood markers in human breast cancer (HBC).

Canine mammary tumors (CMTs) are the most diagnosed neoplasms in dogs; however, there are few studies analyzing the influence of epidemiological, clinicopathological, and histopathological data on cancer-specific survival (CSS), disease-free interval (DFI), and overall survival (OS) in a large cohort. To contribute to the understanding of the biological behavior of this neoplasm, 385 cases were analyzed, 89% malignant, 4% benign, and 7% non-neoplastic lesions. Among the dogs diagnosed with malignant neoplasms, 86% had early clinical stages (I-III), while 14% had regional or distant metastasis at the time of diagnosis. Carcinoma in a mixed tumor was the most frequent histological type with 44% of the cases and had the best prognosis. Analyzed factors such as the presence of pseudocyesis, previous history of the disease, advanced clinical stage (IV-V), and presence of ulceration obtained significant results for CSS, DFI, and OS through univariate analysis and had a negative impact on the survival of the patients. Multivariate analysis showed that histological grading and age proved to be the best independent parameters for the prognostic evaluation of CSS and DFI in this study. These factors were also significant in the overall survival analysis. Therefore, these parameters should be considered valuable risk and prognostic factors for CMTs.

Background: Plasmodium spp. infection triggers the production of inflammatory cytokines that are essential for parasite control, and conversely responsible for symptoms of malaria. Monocytes play a role in host defense against Plasmodium vivax infection and represent the main source of inflammatory cytokines and reactive oxygen species. The anti-inflammatory cytokine IL-10 is a key regulator preventing exacerbated inflammatory responses. Studies suggested that different clinical presentations of malaria are strongly associated with an imbalance in the production of inflammatory and anti-inflammatory cytokines. Methods: A convenience sampling of peripheral blood mononuclear cells from Plasmodium vivax-infected patients and healthy donors were tested for the characterization of cytokine and adenosine production and the expression of ectonucleotidases and purinergic receptors. Results: Here we show that despite a strong inflammatory response, monocytes also bear a modulatory role during malaria. High levels of IL-10 are produced during P. vivax infection and its production can be triggered in monocytes by P. vivax-infected reticulocytes. Monocytes express high levels of ectonucleotidases, indicating their important role in extracellular ATP modulation and consequently in adenosine production. Plasmatic levels of adenosine are not altered in patients experiencing acute malaria; however, their monocyte subsets displayed an increased expression of P1 purinergic receptors. In addition, adenosine decreases Tumor Necrosis Factor production by monocytes, which was partially abolished with the blockage of the A2a receptor. Conclusion: Monocytes have a dual role, attempting to control both the P. vivax infection and the inflammatory response. Purinergic receptor modulators emerge as an untapped approach to ameliorate clinical malaria.

Objetivo: Identificar se os idosos da cidade de Cruzeiro recebem, entendem e seguem as orientações dadas pelos Enfermeiros da Atenção Básica sobre a prevenção da Hipertensão Arterial. Método: O trabalho consiste em uma pesquisa quali-quantitativa, na qual 50 idosos foram questionados sobre seu conhecimento acerca da hipertensão, orientações dos enfermeiros e características da doença. O questionário era composto por 15 perguntas. A pesquisa foi realizada na cidade de Cruzeiro SP, em Unidades de Atenção Básica, num período de janeiro a março de 2022. Resultado: Mesmo com todas as orientações sobre a hipertensão, 88% dos entrevistados não pratica exercícios físicos, 46% sentem falta de ar e 10% não estão tomando os medicamentos corretamente. Conclusão: Pôde-se identificar que, nas unidades investigadas, os enfermeiros passam as informações corretas aos seus pacientes, em relação aos horários e dosagem dos medicamentos, assim como, informam sobre a necessidade de uma alimentação e hidratação adequadas. Porém, os idosos, muitas vezes por esquecimento, deixam de fazer o que lhes foi recomendado.(AU)


Objective: To identify whether the elderly in the city of Cruzeiro receive, understand and follow the guidelines given by Primary Care Nurses on the prevention of Arterial Hypertension. Method: The work consists of a quali-quantitative research, in which 50 elderly people were asked about their knowledge about hypertension, nurses' guidelines and characteristics of the disease. The questionnaire consisted of 15 questions. The survey was carried out in the city of Cruzeiro SP, in Primary Care Units, from January to March 2022. Result: Even with all the guidelines on hypertension, 88% of respondents do not practice physical exercises, 46% feel a lack of air and 10% are not taking their medication correctly. Conclusion: It was possible to identify that, in the investigated units, the nurses pass the correct information to their patients, in relation to the times and dosage of the medicines, as well as, they inform about the need for adequate food and hydration. However, the elderly, often due to forgetfulness, fail to do what was recommended.(AU)

Método: El trabajo consiste en una investigación cuali-cuantitativa, en la que se preguntó a 50 ancianos acerca de su conocimiento sobre hipertensión arterial, directrices de enfermería y características de la enfermedad. El cuestionario constaba de 15 preguntas. La encuesta fue realizada en la ciudad de Cruzeiro SP, en Unidades de Atención Básica, de enero a marzo de 2022. Resultado: Mismo con todas las orientaciones sobre la hipertensión, 88% de los encuestados no practican ejercicios físicos, 46% sienten falta de aire y el 10% no está tomando su medicación correctamente. Conclusión: Fue posible identificar que, en las unidades investigadas, los enfermeros pasan la información correcta a sus pacientes, en relación a los tiempos y dosis de los medicamentos, así como informan sobre la necesidad de alimentación e hidratación adecuadas. Sin embargo, los ancianos, muchas veces por olvidos, dejan de hacer lo recomendado.(AU)

BACKGROUND: The saliva of sand flies, vectors of Leishmania parasites, contains several components that exert pharmacological activity facilitating the acquisition of blood by the insect and contributing to the establishment of infection. Previously, we demonstrated that PpSP32 is the immunodominant salivary antigen in humans exposed to Phlebotomus papatasi bites and validated its usefulness as a predictive biomarker of disease. PpSP32, whose functions are little known to date, is an intriguing protein due to its involvement in the etiopathogenesis of pemphigus, an auto-immune disease. Herein, we aimed to better decipher its role through the screening of several immunomodulatory activity either on lymphocytes or on monocytes/macrophages. METHODS: Peripheral mononuclear cells from healthy volunteers were stimulated with anti-CD3/anti-CD28 antibodies, phytohemagglutinin, phorbol 12-myristate 13-acetate/ionomycin, or lipopolysaccharide in the presence of increasing doses of PpSP32. Cell proliferation was measured after the addition of tritiated thymidine. Monocyte activation was tested by analyzing the expression of CD86 and HLA-DR molecules by flow cytometry. Cytokine production was analyzed in culture supernatants by ELISA. THP-1-derived macrophages were stimulated with LPS in the presence of increasing doses of PpSP32, and cytokine production was analyzed in culture supernatants by ELISA and multiplex technique. The effect of PpSP32 on NF-kB signaling was tested by Western blot. The anti-inflammatory activity of PpSP32 was assessed in vivo in an experimental inflammatory model of carrageenan-induced paw edema in rats. RESULTS: Our data showed that PpSP32 down-modulated the expression of activation markers in LPS-stimulated monocytes and THP1-derived macrophages. This protein negatively modulated the secretion of Th1 and Th2 cytokines by human lymphocytes as well as pro-inflammatory cytokines by monocytes, and THP1-derived macrophages. PpSP32 treatment led to a dose-dependent reduction of IκB phosphorylation. When PpSP32 was injected into the paw of carrageenan-injected rats, edema was significantly reduced. CONCLUSIONS: Our data indicates that PpSP32 induces a potent immunomodulatory effect on monocytes and THP-1-derived macrophages. This inhibition could be mediated, among others, by the modulation of the NF-kB signaling pathway. The anti-inflammatory activity of PpSP32 was confirmed in vivo in the carrageenan-induced paw edema model in rats.

Objetivo: Identificar e conhecer a visão das parturientes em relação ao papel do enfermeiro no parto humanizado. Método:Pesquisa de campo qualiquantitativo de um município do interior do estado de São Paulo. Os dados quantitativos foram tabulados no Excel, e os qualitativos analisados pela temática de Minayo. Resultado: De acordo com os dados obtidos na pesquisa14,28% das participantes reportaram que houve ameaça, foram impossibilitadas de caminhar, procurar posições mais confortáveis e aplicação de episiotomia, a presença do acompanhante foi impossibilitada em 28,6% das participantes. Os toques vaginais por diferentes pessoas aconteceram em 57,14% das participantes, 35,71% relataram o encaminhamento do bebê para sala de procedimentos sem nenhuma justificativa considerável. Conclusão: O papel do enfermeiro é de grande importância para preparar a mulher antes, durante e após o trabalho de parto com orientações sobre os seus direitos,o enfermeiro deve estar preparado constantemente para atender a sua comunidade(AU)

Abstract Background The many combinations of chemotherapeutic agents and biologicals available in the Brazilian National Health System for the treatment of metastatic breast cancer require analysis that contribute to decision making. Objective The study's primary aim was to evaluate the first-line treatment of HER2- overexpressing metastatic breast cancer from the Brazilian Unified Health System perspective using multicriteria decision analysis (MCDA). Method The treatment options evaluated were (a) pertuzumab combined with trastuzumab and docetaxel, and (b) trastuzumab in combination with docetaxel. Using the hierarchical analytical method, medical oncologists compared the relevance of five predefined criteria: overall survival, response to treatment, adverse events, cost- effectiveness, and budget impact. Results The therapeutic scheme considered more appropriate by the model was pertuzumab combined with trastuzumab and docetaxel. The most sensitive criteria were adverse events, cost-effectiveness, and budget impact. The results suggest that the classification has a close relationship with the perspective of healthcare professionals participating in the questionnaire. Conclusion Defining the treatment of an incurable disease associated with a short survival time and high-cost treatment options necessitates complex decision-making. MCDA allows the weighting of criteria and considering criteria that would be difficult to measure in other methods, such as cost-effectiveness. These aspects differ from economic models and contribute to a broader evaluation of health decision-making.


Resumo Introdução As diversas combinações de agentes quimioterápicos e biológicos disponíveis no Sistema Único de Saúde brasileiro para o tratamento do câncer de mama metastático requerem análises que contribuam para a tomada de decisões. Objetivo O objetivo principal deste estudo foi avaliar o tratamento de primeira linha para câncer de mama metastático HER2 hiperexpresso sob a perspectiva do Sistema Único de Saúde, utilizando a análise de decisão multicritérios (MCDA). Método As opções de tratamento avaliadas foram: (a) pertuzumabe em combinação com trastuzumabe e docetaxel, e (b) trastuzumabe em combinação com docetaxel. Usando o método analítico hierárquico, médicos oncologistas compararam a relevância dos cinco critérios predefinidos: sobrevida global, resposta ao tratamento, eventos adversos, custo-efetividade e impacto orçamentário. Resultados O esquema terapêutico considerado mais apropriado pelo modelo foi pertuzumabe em combinação com trastuzumabe e docetaxel. Os critérios mais sensíveis foram eventos adversos, custo-efetividade e impacto orçamentário. Os resultados sugerem que a classificação está associada à perspectiva do profissional de saúde participante do questionário. Conclusão Definir o tratamento de uma doença incurável associada a um tempo de sobrevida curto e opções de tratamento de alto custo requer uma tomada de decisão complexa. O MCDA permite ponderar e considerar critérios que seriam difíceis de medir em outros modelos de decisão. Esses aspectos contribuem para uma avaliação mais ampla da tomada de decisões em saúde.

Objetivo: identificar a percepção da equipe de enfermagem atuante no Serviço de Atendimento Móvel de Urgência acerca das competências forenses. Método: estudo de natureza descritivo-exploratório com abordagem qualitativa,realizado com 11 profissionais que atuam em Serviço de Atendimento Móvel de Urgência. Os dados foram interpretados à luz do referencial teórico segundo Bardin. Resultado: emergiram quatro categorias centrais e uma subcategoria: Percepção da equipe de enfermagem acerca da singularidade da Enfermagem Forense; Sapiência dos participantes no que concerne à especialização da Enfermagem Forense; Experiências e vivências frente ao processo de preservação dos vestígios e evidências durante o resgate em situação forenses; Enfrentamento diante de ocorrências forenses; Vulnerabilidade, sentimentos e lembranças vivenciadas em ocasiões forenses. Conclusão: evidenciou-se a ausência da capacitação dos profissionais envolvidos nesse cenário. Percebeu-se, ainda, durante a análise das respostas dadas, que os participantes conhecem parcialmente o que é, qual a aplicabilidade e quais as competências da enfermagem forense.(AU)


Objective: to identify the perception of the nursing team working in the Mobile Emergency Care Service about forensic competences Method: a descriptive-exploratory study with a qualitative approach, carried out with 11 professionals who work in the Mobile Emergency Care Service. The data were interpreted in the light of the theoretical framework according to Bardin. Result: four central categories and a subcategory emerged: Perception of the nursing team about the uniqueness of Forensic Nursing; Participants' sapience regarding the specialization of Forensic Nursing; Experiences and experiences in the process of preserving traces and evidence during the rescue in forensic situations; Coping with forensic events; Vulnerability, feelings and memories experienced in forensic occasions. Conclusion: the lack of training of professionals involved in this scenario was evidenced. It was also noticed, during the analysis of the answers given, that the participants partially know what it is, what is the applicability and what are the competences of forensic nursing.(AU)

Resultado: surgieron cuatro categorías centrales y una subcategoría: Percepción del equipo de enfermería sobre la singularidad de la Enfermería Forense; Sabiduría de los participantes sobre la especialización en Enfermería Forense; Vivencias y vivencias en el proceso de preservación de huellas y evidencias durante el rescate en situaciones forenses; Afrontamiento de eventos forenses; Vulnerabilidad, sentimientos y recuerdos vividos en ocasiones forenses. Conclusión: se evidenció la falta de formación de los profesionales involucrados en este escenario. También se constató, durante el análisis de las respuestas dadas, que los participantes conocen parcialmente qué es, cuál es la aplicabilidad y cuáles son las competencias de la enfermería forense. RESUMO

The development of 3D-printed electrochemical sensors by fused deposition modeling (FDM) has been increasing exponentially in the last five years. In this context, commercial conductive filaments composed of a blend of carbon particles (e.g., graphene or carbon black (CB)) and insulating thermoplastic polymers (e.g., polylactic acid (PLA) or acrylonitrile butadiene styrene (ABS)) have been widely used for electrode fabrication. However, such materials may be expensive and the electrodes when used "as-printed" exhibit poor electrochemical performance as a function of the low content of conductive particles in the composition (∼10 to 20 wt%), which requires one or more post-treatment steps (e.g. polishing, chemical, electrochemical, and photochemical) to reach good electrochemical performance. In this technical note a less used approach to produce "ready-to-use" electrochemical platforms based on 3D printing is explored, which consists of the coating of 3D-printed insulating substrates with homemade conductive composites. To demonstrate the potentiality of this alternative protocol, 3D-printed ABS insulating substrates at two geometries were coated in a highly loaded graphite (55 wt%) homemade composite (G-ABS) and evaluated for the detection of the ferri/ferrocyanide redox probe and model analytes in stationary and hydrodynamic 3D-printed systems (nitrite in micro-flow injection analysis/µFIA and paracetamol in batch injection analysis/BIA, respectively). The analytical parameters acquired with the coated electrodes were comparable to those obtained using conventional electrodes (glassy carbon, boron-doped diamond and carbon screen-printed) and 3D-printed sensors fabricated with commercial filaments. Moreover, the inclusion of carbon black in the fluid conductive composite was demonstrated as a perspective to obtain modified coated 3D-printed surfaces easily for the first time. This alternative "do it yourself" strategy is promising for the large-scale production of very cheap (US$ 0.08) and high-performance electrodes based on FDM 3D printing. Moreover, this approach dispenses the acquisition of commercial conductive filaments and the laborious development of homemade filaments.

Cutaneous round cell neoplasms (RCNs) are among the most common skin tumors of dogs. Although a diagnosis can be made in most cases following the examination of H&E-stained routine samples, less-differentiated neoplasms can exhibit overlapping morphologic features that may be challenging to the pathologist, and require immunohistochemistry or molecular testing to reach a final diagnosis. Many patients with cutaneous RCNs are initially seen by a general practitioner and are not referred to a veterinary teaching hospital until a diagnosis has been made. For this reason, anatomic pathology residents at some academic or other training institutions may not be exposed frequently to RCNs during the surgical biopsy service as part of their residency training. In an attempt to fill that gap, here we review the key routine histologic features of canine cutaneous RCNs.

Introduction: Background: we aimed to investigate the isolated effect of tele-exercises (TE) and their combined effect with nutritional coaching (NC) on health-related parameters of overweight and obese individuals. Methods: forty-one overweight (body mass index ≥ 25 kg/m2) and obese (body mass index ≥ 30 kg/m2) women were randomly assigned to the experimental groups: TE (n = 20) or TE+NC (n = 21). TE was applied 3 days/week in both groups, while TE+NC also received NC 1 day/week. Anthropometric, body composition, and exercise capacity-related outcomes, quality of life, and eating behavior were assessed before and after 8 weeks of the intervention. Results: a significant main time effect (p < 0.01) was detected for flexibility, isometric muscle strength and dynamic muscle endurance, but no main group effect was noted (p > 0.05). On the other hand, neither a significant main time nor group effect (p > 0.05) was detected in the anthropometric and body composition measures, quality of life, or eating behavior. Similarly, no significant between-group difference was observed in the absolute or relative change analysis (all comparisons, p > 0.05). Conclusions: an 8-week TE program enhanced exercise capacity, but did not impact anthropometric or body composition-related outcomes. The combination of NC+TE did not have a clinical advantage in the management of overweight and obesity.


Introducción: Introducción: nuestro objetivo fue investigar el efecto aislado de los tele-ejercicios (TE) y su efecto combinado con el coaching nutricional (CN) sobre los parámetros relacionados con la salud de las personas con sobrepeso y obesidad. Métodos: cuarenta y una mujeres con sobrepeso (índice de masa corporal ≥ 25 kg/m2) y obesas (índice de masa corporal ≥ 30 kg/m2) fueron asignadas aleatoriamente a los grupos experimentales: TE (n = 20) o TE+CN (n = 21). La TE se aplicó 3 días/semana en ambos grupos, mientras que el grupo TE+CN también recibió NC 1 día/semana. Se evaluaron los resultados antropométricos, la composición corporal y la capacidad de ejercicio, la calidad de vida y la conducta alimentaria antes y después de 8 semanas de intervención. Resultados: se detectó un efecto de tiempo principal significativo (p < 0,01) para la flexibilidad, la fuerza muscular isométrica y la resistencia muscular dinámica, pero no se observó ningún efecto de grupo principal (p > 0,05). Por otro lado, no se detectó ningún efecto de tiempo principal ni de grupo significativo (p > 0,05) en las medidas antropométricas y de composición corporal, calidad de vida o conducta alimentaria. De manera similar, no se observaron diferencias significativas entre los grupos en el análisis del cambio absoluto o relativo (todas las comparaciones, p > 0,05). Conclusiones: un programa de TE de 8 semanas mejoró la capacidad de ejercicio pero no afectó los resultados antropométricos o relacionados con la composición corporal. La combinación de CN + TE no obtuvo ninguna ventaja clínica en el manejo del sobrepeso y la obesidad.

SUMMARY: Previous evidence indicates that body fat can distinguish Brazilian jiu-jitsu (BJJ) athletes according to the competitive level. However, propositions of cut-off points for establishing classifications of body fat percentage for combat sports athletes and, specifically, for BJJ athletes are still incipient in the literature. In this sense, the main aim of the present study was to establish a normative table for the classification of body fat percentage in BJJ athletes. As a secondary aim, athletes were compared according to competitive level. Ninety male BJJ athletes (aged: 29.0 ± 8.2 years; practice time: 6.0 ± 2.1 years; body mass: 82.1 ± 12.7 kg; height: 175.9 ± 6.5 cm; fat mass: 16.0 ± 8.9 kg; bone mineral content: 3.7 ± 0.6 kg; muscle mass: 37.9 ± 5.4 kg; body fat percentage: 17.3 ± 6.8 %; basal metabolic rate: 1811.4 ± 193.4 kcal) from different competitive levels: state (n= 42), national (n= 26) and international (n= 22) took part in this study. All athletes had their body composition measured via tetrapolar bioelectrical impedance. Percentiles p10, p25, p50, p75, and p90 were used to establish the classification. As a result, the following classification was obtained: <7.7 % (very low); ≥7.7-11.5 % (low); 11.6-17.0 % (medium); 17.1-24.0 % (high) and ≥24.1 % (very high). State-level athletes had a higher fat percentage than national and international-level athletes (p<0.05). The proposed cut-off points can help professionals responsible for sports training and nutritional prescription in monitoring the body fat of BJJ athletes.


RESUMEN: La grasa corporal puede distinguir a los atletas de jiu-jitsu brasileño (BJJ) según el nivel competitivo. Sin embargo, las propuestas de puntos de corte para establecer clasificaciones de porcentaje de grasa corporal para deportistas de deportes de combate y, específicamente, para deportistas de BJJ son aún incipientes en la literatura. En este sentido, el objetivo principal del presente estudio fue establecer una tabla normativa para la clasificación del porcentaje de grasa corporal en atletas de BJJ. Secundariamente, comparar a los deportistas según el nivel competitivo. Participaron noventa atletas masculinos de BJJ (edad: 29,0 ± 8,2 años; tiempo de práctica: 6,0 ± 2,1 años; masa corporal: 82,1 ± 12,7 kg; estatura: 175,9 ± 6,5 cm; masa adiposa: 16,0 ± 8,9 kg; contenido mineral óseo: 3,7 ± 0,6 kg; masa muscular: 37,9 ± 5,4 kg; porcentaje de grasa corporal: 17,3 ± 6,8 %; tasa metabólica basal: 1811,4 ± 193,4 kcal) de diferentes niveles competitivos: estatal (n= 42), nacional (n= 26) e internacional (n= 22). Se evaluó la composición corporal a través de bioimpedancia tetrapolar. Para establecer la clasificación se utilizaron los percentiles p10, p25, p50, p75 y p90. Como resultado se obtuvo la siguiente clasificación: <7,7 % (muy bajo); ≥7,7-11,5 % (bajo); 11,6-17,0 % (medio); 17,1-24,0 % (alto) y ≥24,1 % (muy alto). Los atletas de nivel estatal tenían un porcentaje de grasa corporal más alto respecto a los atletas de nivel nacional e internacional (p <0.05). Los puntos de corte propuestos pueden ser útiles para monitorizar la grasa corporal de los atletas de BJJ.

Backgroung: The World Health Organization (WHO) recommends the use of the body mass index (BMI) as a cost-effective method to assess the nutritional status at the population level. The increase of BMI is linked to a higher risk of other chronic non-communicable diseases (NCDs), particularly hypertension, type 2 diabetes mellitus (T2DM), dyslipidemias, and some types of cancer. The prevalence of obesity has increased worldwide, and this condition has dramatically affected children and adolescents. Obesity at a young age increases the chances of severe obesity and its complications in adults.Objective: This study aimed to establish cut-off points for body fat percentage in male and female adolescents aged 16 to 18 years using bioelectrical impedance (InBody 570®). Methods: Gender specific tables were proposed based on the percentiles 3, 5, 10, 15, 25, 50, 75, 85, 95 and 97. A total of 546 adolescents were included. Results: The body fat percentage cut-off points for the male group were: P3 = 6.0-7.0%; P5 = 7.1-8.9%; P10 = 9.0-9.8%; P15 = 9.9-11.7%; P25 = 11.8-15.5%; P50 = 15.6-21.9%; P75 = 22.0-27.8%; P85 = 27.9-36.0%; P95 = 36.1-38.0% and P97 ≥ 38.1%. For females, the cut-off points were: P3 = 9.5-10.0%; P5 = 10.1-11.0%; P10 = 11.1-11.8%; P15 = 11.9-14.0%; P25 = 14.1-19.0%; P50 = 19.1-27.1%; P75 = 27.2-29.0%; P85 = 29.1-39.9%; P95 = 40.0-51.0% and P97 ≥ 51.0%. Conclusion: The establishment of cut-off points for body fat percentage may improve the clinical assessment and management of overweight and obese adolescents.


Introdução: A Organização Mundial da Saúde (OMS) recomenda o uso do índice de massa corporal (IMC) como método custo-efetivo nível 1 para avaliar o estado nutricional na população. O aumento do IMC está associado a um maior risco de outras doenças crônicas não transmissíveis (DCNT), particularmente a hipertensão arterial sistêmica (HAS), diabetes mellitus tipo 2 (DM2), dislipidemias e alguns tipos de cânceres. A prevalência da obesidade tem aumentado em todo o mundo e essa condição tem afetado dramaticamente crianças e adolescentes. A obesidade em jovens, por sua vez, aumenta as chances de obesidade grave e suas complicações em adultos.Objetivo: Este estudo teve como objetivo estabelecer pontos de corte para o percentual de gordura corporal em adolescentes do sexo masculino e feminino de 16 a 18 anos, utilizando a bioimpedância elétrica (InBody 570®). Método: Tabelas específicas para o sexo masculino e feminino foram propostas, com base nos percentis 3, 5, 10, 15, 25, 50, 75, 85, 95 e 97. Foram incluídos 546 adolescentes. Resultados: Os pontos de corte do percentual de gordura corporal para o grupo masculino foram: P3 = 6,0-7,0%; P5 = 7,1-8,9%; P10 = 9,0-9,8%; P15 = 9,9-11,7%; P25 = 11,8-15,5%; P50 = 15,6-21,9%; P75 = 22,0-27,8%; P85 = 27,9-36,0%; P95 = 36,1-38,0% and P97 ≥ 38,1%. Introducción: El sobrepeso afecta negativamente al sistema cardiovascular y al osteomioarticular, favoreciendo el dolor en las articulaciones de las extremidades inferiores. Por lo tanto, se han aplicado estrategias para reducir el sobrepeso a través de cambios en el estilo de vida. Objetivo: investigar los efectos de dos programas de intervención multidisciplinares sobre la aptitud física relacionada con la salud de mujeres obesas con síntomas de dolor en las rodillas. Métodos: Se evaluó a mujeres entre 40 y 59 años con obesidad (n=59) en el tiempo pre y post intervención en antropometría, composición corporal, aptitud física, nivel de discapacidad en rodilla y cadera y calidad de vida relacionada con la salud. Se realizó un ensayo clínico aleatorizado de grupos paralelos, utilizando dos programas de ejercicio de resistencia y reeducación alimentaria, durante ocho semanas. Todos los participantes recibieron el mismo contenido de intervención nutricional y para ejercicios físicos se aleatorizaron en dos grupos, G1 siendo el "grupo de fuerza" utilizando máquinas (n=30), y G2 siendo el grupo de "entrenamiento funcional" (n=29). Veintiocho mujeres (G1=18 + G2=10) fueron incluidas en los análisis finales. Los grupos y el tiempo se evaluaron mediante análisis bidireccional de la varianza, con medidas repetidas. Se asumió una p < 0,05 para todos los análisis. Resultados: se identificó un efecto de tiempo, con valores más altos para la fuerza máxima de tracción lumbar isométrica (MILTS), luego del período de intervención en ambos grupos (p < 0.05). Sin embargo, para las otras variables investigadas, no se identificaron diferencias significativas (p > 0.05). Conclusión: La ausencia de resultados significativos para todas las variables, excepto FIMTL, puede estar justificada por la adherencia moderada de los participantes a las intervenciones (~ 59% a 66%) ya que el estudio se realizó durante el período de aislamiento social (COVID-19). Sin embargo, se cree que los modelos de intervención fueron positivos en el mantenimiento de las variables estudiadas.


Introduction: Overweight negatively impacts the cardiovascular system and osteomyoarticular, promoting pain in the joints of the lower limbs. Therefore, strategies have been applied to reduce overweight through lifestyle changes. Objective: to investigate the effects of two multidisciplinary intervention programs on the health-related physical fitness of obese women with pain symptoms in the knees. Methods: Women between 40 and 59 years of age with obesity (n=59) were evaluated in the pre- and post-intervention time regarding anthropometry, body composition, physical fitness, level of disability in the knee and hip, and health-related quality of life. A randomized clinical trial of parallel groups was conducted, using two resistance exercise programs and food re-education, for eight weeks. All participants received the same nutritional intervention content and for physical exercises were randomized into two groups, G1 being the "strength group" using machines (n=30), and G2 being the "functional training" group (n=29). Twenty-eight women (G1=18 + G2=10) were included in the final analyses. The groups and time were evaluated by two-way analysis of variance, with repeated measures. A p < 0.05 was assumed for all analyses. Results: a time effect was identified, with higher values for maximum isometric lumbar-traction strength (MILTS), after the intervention period in both groups (p < 0.05). However, for the other variables investigated, no significant differences were identified (p > 0.05). Conclusion: The absence of significant results for all variables, except FIMTL, may be justified by the moderate adherence of the participants to interventions (~59% to 66%) since the study was conducted during the period of social isolation (COVID-19). However, it is believed that the intervention models were positive in maintaining the variables studied.

Visceral leishmaniasis (VL) is the most severe clinical form of leishmaniasis, fatal if untreated. Vaccination is the most cost-effective approach to disease control; however, to date, no vaccines against human VL have been made available. This work examines the efficacy of a novel vaccine consisting of the Leishmania membrane protein KMP11, LEISH-F3+ (a recombinant fusion protein, composed of epitopes of the parasite proteins nucleoside hydrolase, sterol-24-c-methyltransferase, and cysteine protease B), and the sand fly salivary protein LJL143, in two dose ratios. The inclusion of the TLR4 agonist GLA-SE as an adjuvant, and the use of virosomes (VS) as a delivery system, are also examined. In a hamster model of VL, the vaccine elicited antigen-specific immune responses prior to infection with Leishmania infantum. Of note, the responses were greater when higher doses of KMP11 and LEISH-F3+ proteins were administered along with the GLA-SE adjuvant and/or when delivered within VS. Remarkably, hamsters immunized with the complete combination (i.e., all antigens in VS + GLA-SE) showed significantly lower parasite burdens in the spleen compared to those in control animals. This protection was underpinned by a more intense, specific humoral response against the KMP11, LEISH-F3+, and LJL143 antigens in vaccinated animals, but a significantly less intense antibody response to the pool of soluble Leishmania antigens (SLA). Overall, these results indicate that this innovative vaccine formulation confers protection against L. infantum infection, supporting the advancement of the vaccine formulation into process development and manufacturing and the conduction of toxicity studies towards future phase I human clinical trials.

Objetivo: Desvelar as fragilidades e a vivência de enfermeiros na abordagem de família do doador de órgãos e tecidos. Método: Pesquisa de abordagem qualitativa com análise de conteúdo de Bardin, realizada com sete enfermeiros em um hospital de médio porte no interior de São Paulo, Brasil. Resultado: Os dados obtidos foram agrupados em três categorias centrais: Conhecimento dos enfermeiros no contexto da abordagem familiar do potencial doador de órgãos e tecidos; Fragilidades vivenciadas por enfermeiros no contexto da comunicação de más notícias para familiares do potencial doador de órgãos e tecidos e Experiências de enfermeiros diante do diagnóstico de morte encefálica e dos fatores determinantes para obtenção de órgãos e tecidos. Conclusão: As experiências dos enfermeiros frente, às fragilidades encontradas e vivenciadas na abordagem familiar de potencial doador de órgãos, são caracterizadas por conflitos e situações que expõem o profissional a uma atmosfera de sentimentos de acolhimento e de empatia.(AU)


Objective: To unveil the weaknesses and the experience of nurses in approaching the organ and tissue donor family. Method: Qualitative research with content analysis by Bardin, carried out with seven nurses in a medium-sized hospital in the interior of São Paulo, Brazil. Results: The data obtained were grouped into three central categories: Nurses' knowledge in the context of the family approach to the potential organ and tissue donor; Weaknesses experienced by nurses in the context of communicating bad news to family members of the potential organ and tissue donor and Nurses' experiences in the face of brain death diagnosis and the determining factors for obtaining organs and tissues. Conclusion: The nurses' experiences in face of the weaknesses found and experienced in the family approach of potential organ donors, are characterized by conflicts and situations that expose the professional to an atmosphere of feelings of welcome and empathy.(AU)


Objetivo: Dar a conocer las debilidades y la experiencia de las enfermeras en el acercamiento a la familia del donante de órganos y tejidos. Método: Investigación cualitativa con análisis de contenido de Bardin, realizada con siete enfermeras en un hospital de mediano tamaño del interior de São Paulo, Brasil. Resultado: Los datos obtenidos se agruparon en tres categorías centrales: Conocimiento del enfermero en el contexto del abordaje familiar del potencial donante de órganos y tejidos; Debilidades vividas por enfermeras en el contexto de comunicar malas noticias a familiares del potencial donante de órganos y tejidos y Experiencias de enfermeras ante el diagnóstico de muerte encefálica y los determinantes para la obtención de órganos y tejidos. Conclusión: Las vivencias de las enfermeras ante las debilidades encontradas y vividas en el abordaje familiar de los potenciales donantes de órganos, se caracterizan por conflictos y situaciones que exponen al profesional a un clima de sentimientos de acogida y empatía.(AU)

INTRODUCTION: Investigating the of overweight or obese adolescents' nutritional quality is substantial to guide healthy eating strategiesOBJECTIVE: To assess the effects of a multi-professional approach on anthropometric parameters, body composition, and the level of food processing in adolescentsMETHODS: Twelve overweight/obese male adolescents between 13-17 years old participated in a series of 12-week multi-professional interventions led by physical education, nutrition, and psychology professionals. The participants were assessed before the intervention as well as at 6 and 12 weeks. Anthropometric assessments were performed, including body weight, stature, and body mass index; body composition, with fat mass (FM), body fat (%BF), skeletal muscle mass, and resting metabolic rate (RMR), and by filling in a three-day food record, to assess the ingested foods by its level of processing: fresh, minimally processed, processed, and ultra-processedRESULTS: No differences were observed in anthropometric parameters performed at different stages (p>0.05). Reductions in FM and %BF and increases in SMM and RMR (p<0.05) were identified after, the 12-week intervention. There was an increase in the consumption of natural foods/g and increased the caloric consumption of processed foods/kcal after 12-week of intervention (p<0.05CONCLUSION: The model resulted in a decrease in fat mass and body fat percentage, an in skeletal muscle mass, resting metabolic rate, natural foods/g, and processed foods/kcal


INTRODUÇÃO: Investigar a qualidade nutricional de adolescentes com sobrepeso ou obesidade é substancial para orientar estratégias de alimentação saudávelOBJETIVO: Avaliar os efeitos de uma abordagem multiprofissional sobre os parâmetros antropométricos, de composição corporal e o nível de processamento de alimentos em adolescentesMÉTODO: Doze adolescentes do sexo masculino com sobrepeso/obesidade, entre 13-17 anos participaram de uma série de intervenções multiprofissionais ao longo de 12 semanas, que foram conduzidas por profissionais de educação física, nutricionistas e psicólogos. Os participantes foram avaliados antes da intervenção, após 6 e após 12 semanas. As avaliações antropométricas foram realizadas, incluindo o peso corporal, a estatura e o índice de massa corporal; para a composição corporal foram incluídas: a massa gorda (MG), percentual de gordura corporal (%G), massa muscular esquelética (MME) e taxa metabólica basal (TMB), bem como o preenchimento do registro alimentar de três dias, para avaliar os alimentos ingeridos, via nível de processamento: in natura, minimamente processado, processado e ultraprocessadoRESULTADOS: Não foram observadas diferenças significativas nos parâmetros antropométricos realizados nas diferentes etapas (p>0,05). Reduções na MG e %G e aumentos na MME e TMB (p<0,05) foram identificados após 12 semanas de intervenção. Houve um aumento do consumo de alimentos in natura/g e aumento do consumo calórico de alimentos industrializados/kcal, após 12 semanas de intervenção (p<0,05CONCLUSÃO: O modelo proposto resultou em uma diminuição da massa gorda e percentual de gordura corporal, aumento da massa muscular esquelética, taxa metabólica basal, consumo de alimentos naturais/g e alimentos processados/kcal

Monocytes play an important role in the host defense against Plasmodium vivax as the main source of inflammatory cytokines and mitochondrial reactive oxygen species (mROS). Here, we show that monocyte metabolism is altered during human P. vivax malaria, with mitochondria playing a major function in this switch. The process involves a reprograming in which the cells increase glucose uptake and produce ATP via glycolysis instead of oxidative phosphorylation. P. vivax infection results in dysregulated mitochondrial gene expression and in altered membrane potential leading to mROS increase rather than ATP production. When monocytes were incubated with P. vivax-infected reticulocytes, mitochondria colocalized with phagolysosomes containing parasites representing an important source mROS. Importantly, the mitochondrial enzyme superoxide dismutase 2 (SOD2) is simultaneously induced in monocytes from malaria patients. Taken together, the monocyte metabolic reprograming with an increased mROS production may contribute to protective responses against P. vivax while triggering immunomodulatory mechanisms to circumvent tissue damage. IMPORTANCE Plasmodium vivax is the most widely distributed causative agent of human malaria. To achieve parasite control, the human immune system develops a substantial inflammatory response that is also responsible for the symptoms of the disease. Among the cells involved in this response, monocytes play an important role. Here, we show that monocyte metabolism is altered during malaria, with its mitochondria playing a major function in this switch. This change involves a reprograming process in which the cells increase glucose uptake and produce ATP via glycolysis instead of oxidative phosphorylation. The resulting altered mitochondrial membrane potential leads to an increase in mitochondrial reactive oxygen species rather than ATP. These data suggest that agents that change metabolism should be investigated and used with caution during malaria.
